The Co-Transcriptional Capping T7 In Vitro Transcription Reagent Kit has been optimized for efficient transcription reactions. Using T7 RNA Polymerase, the kit enables in vitro transcription from linear double-stranded DNA templates containing a T7 promoter sequence. NTPs serve as substrates, while the capping reagent CAP GAG (3'OMe) is co-transcriptionally incorporated at the 5' end of the mRNA. This allows users to quickly and easily produce large quantities of RNA with Cap1 modification.

The inclusion of Cap1 enhances mRNA stability by protecting it from degradation, ensuring high translation efficiency. The kit also provides a commonly used modified nucleotide, pUTP, which can be optionally incorporated based on specific transcription needs. These modified nucleotides effectively reduce mRNA immunogenicity and suppress innate immune activation, making mRNA a promising tool for regenerative medicine, disease treatment, and cellular reprogramming.

A single reaction with this kit can yield 150–200 μg of RNA, with scalability for milligram-level RNA production. The synthesized RNA can be used for various applications, including RNA structure and function studies, RNase protection assays, probe hybridization, antisense RNA, and RNAi research.
