Product Overview

The T7 High-Temperature RNA Polymerase enables in vitro transcription at elevated temperatures (up to 50°C). Using linear double-stranded DNA with a T7 promoter sequence as the template and NTPs as substrates, the enzyme transcribes downstream DNA sequences to produce RNA products. It can also utilize modified nucleotides to produce RNA labeled with biotin, dyes, or radioactive markers, as well as capped RNA using cap analogs or cap-like structures.

High-temperature reactions help reduce the formation of dsRNA by-products, lowering the immunogenicity of the transcribed RNA. The kit includes commonly used modified nucleotide N1-Me-pUTP for specialized transcription needs. Each reaction can yield 150–200 μg of RNA, with scalability for milligram-level RNA production.

The transcribed RNA is suitable for various applications, such as RNA structure and function studies, RNase protection assays, probe hybridization, anti-sense RNA, and RNAi. It can also be capped using the Vaccinia Virus Capping System (HBP000606) and 2'-O-Methyltransferase (HBP000701), or polyadenylated using Poly(A) Polymerase (HBP000801) to produce mRNA for downstream applications like in vitro translation or transfection.
