The T7 In Vitro Transcription Kit (Low dsRNA) is designed to produce large amounts of RNA with reduced double-stranded RNA (dsRNA) by-products. This kit leverages a mutant T7 RNA polymerase and an optimized transcription system to efficiently transcribe RNA from linear double-stranded DNA templates containing a T7 promoter sequence, using NTPs as substrates.

The kit also supports transcription using modified nucleotides to produce RNA labeled with biotin, dyes, or radioactive markers, as well as capped RNA using cap analogs or cap-like structures. It includes the commonly used modified nucleotide N1-Me-pUTP to meet specialized transcription needs. Each reaction yields 150–200 μg of RNA and can be scaled up to produce milligram-level quantities.

The transcribed RNA is suitable for various applications, including RNA structure and function studies, RNase protection assays, probe hybridization, anti-sense RNA, and RNAi. Additionally, it supports mRNA production via capping using the Vaccinia Virus Capping System (HBP000606) and 2'-O-Methyltransferase (HBP000701), and polyadenylation using Poly(A) Polymerase (HBP000801), for use in downstream applications like in vitro translation and transfection.
