The High-Yield T7 In Vitro Transcription Kit has been optimized for efficient transcription reactions. Using T7 RNA Polymerase, this kit enables in vitro transcription from linear double-stranded DNA templates containing a T7 promoter sequence, with NTPs serving as substrates. It provides users with a simple and fast method to produce large quantities of RNA.

The kit supports the use of modified nucleotides as substrates, allowing for the production of RNA labeled with biotin, dyes, or radioactive markers. It can also generate capped RNA using cap structures or cap analogs as substrates. A commonly used modified nucleotide, N1-Me-pUTP, is included for optional use to meet specific transcription needs.

Each reaction yields 150–200 μg of RNA, and production can be scaled up to milligram levels. The transcribed RNA is suitable for various applications, such as RNA structure and function studies, RNase protection assays, probe hybridization, antisense RNA, and RNAi experiments. Additionally, mRNA with downstream modifications can be produced, including capping with the vaccinia capping system (HBP000606) or 2’-O-methyltransferase (HBP000701), and polyadenylation using Poly(A) Polymerase (HBP000801). These modifications enable mRNA applications in in vitro translation, transfection, and other downstream workflows.
