Introduction
Granulocyte Macrophage Colony Stimulating Factor (GM-CSF) was first characterized as a growth factor that supports the in-vitro colony formation of granulocytes-macrophages progenitor cells. It is a pleiotropic cytokine and a member of a family of endogenous cytokines of the hematopoietic system. GM-CSF is produced as a response to immune or inflammatory stimuli by activated cells of the hematopoietic system such as T cells, B cells, macrophages, mast cells and also fibroblasts and alveolar epithelial cells. It plays an important role in regulating the proliferation, differentiation, survival and activation of hematopoietic cells such as granulocytes and monocytes, neutrophiles, basophiles and eosonophoiles, erythroid cells, megakaryocytes, and T cells. Bacterial endotoxins, TNF, Interleukin-1, and GM-CSF induce the synthesis of G-CSF.

Recombinant human Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF) protein is expressed with sequence (A18-E144) of human GM-CSF. It is a 14.6 kDa globular protein consisting of 128 amino acids (with the N-terminal methionine) containing two intramolecular disulfide bonds and two potential N-linked glycosylation sites.
N-terminal sequence analysis: (Met)-Ala-Pro-Ala-Arg-Ser-Pro-Ser-Pro-Ser-Thr-Gln-Pro-Trp-Glu-His.
The ED50 as determined by the dose-dependant stimulation of the proliferation of human TF-1 cells (human erythroleukemic indicator cell line) is less than 0.1 ng/ml, corresponding to a specific activity of 1.0 x 10^7 IU/mg.
Purity: > 95% by SEC-HPLC and reducing SDS-PAGE.
Endotoxin: < 0.01 ng/ug (0.1 IEU/ug).
Formulation: Lyophilized from a 0.2 um filtered solution of 20 mM PB, 150 M NaCl, 5% Mannitol, pH 7.2.
Reconstitution: Always centrifuge tubes before opening. Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100 ug/ml. Dissolve the lyophilized protein in 1X PBS. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
