Background

In vivo grade recombinant natural and engineered mouse IgG1 Fc proteins (mIgG1 Fc), and recombinant mouse IgG1 isotype control antibody and mutants are available. Biotinylated mIgG1 Fc, and mIgG1 Fc with Avi-, His-, and Flag-tags are also available. The recombinant mouse IgG1 Fc protein and mutants can be used as the isotype control of the mIgG1 antibodies and the mIgG1 Fc fusion proteins in various in vitro and in vivo experiments.

The T252M substitution of the mIgG1 Fc protein can significantly increase the affinity to protein A.

The in vivo grade recombinant mouse IgG1 Fc protein (mIgG1 Fc, Val 98 - Lys 324 of the constant region of the IgG1 heavy chain, Accession # P01868-1) was produced in mammalian cells and purified by protein G. The mIgG1 Fc protein includes the CH2 (111-217) and CH3 (218-324) domains of the heavy chain constant region as well as the hinge region (98-110) before the CH2 domain. The C102S version of the mIgG1 Fc protein is also available. The C102S substitution prevents improper cysteine bond formation in the absence of a light chain.
Molecular weight: Predicted 25.8 kDa. Due to glycosylation, the recombinant mouse IgG1 Fc protein migrates to 32-35 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.
