Description:

Q5 DT™ 2x Sequencing Mix provides premixed reagents for Sanger sequencing reactions. It is a perfect alternative for BigDye™. It has been formulated to get long high quality read with uniform peak height, and to read through complex sequencing structure. The Q5 DT™ 2x ReadySeq Sequencing Mix contains all required reagent components for the sequencing reaction in a ready reaction, pre-mixed format.

Standard Protocol:

01 Assembly Sequencing Reaction

Set up 10ul sequencing reaction

Plasmid 150-200 ng

PCR 10ng/kb

Primer:3-5uM, 1ul

Add H2O to final Volume of 5 ul

Add 5 ul 2x ReadySeq Mix

02 Perform Cycle Sequencing

Place the tubes or plates in a thermal cycler and run

the following PCR cycles:

96ºC 2 min, 1 cycle

96ºC 10 sec, 50ºC 5sec, 60ºC 4min, 25cycles

4ºC hold

03 Post Sequencing Clean up

Any post sequencing clean up protocol

04 Capillary Electrophoresis
